If you don't do the Mages Guild then you won't be able to get into the Arcane University... and you won't get to make cool enchanted armor and spells

Yes, there should be a guy sweeping outside that you talk to, to get the key. This place is quest related though... you get the quest from the scribe at the jefferson memorial assuming you finished the main quest of the original game...
