Mass Effect, but since this is the PS3 forum, Gran Turismo 5. I know it's been delayed from a March release in Japan to "TBA", but it's still my most anticipated game this year. I fully expect to spend over a thousand hours playing GT5 before GT6 comes out.

I've spent hundreds of hours playing Counter-Strike and Diablo II in my lifetime, about 170 hours on CoD4 online, and I've probably beaten FFVII a dozen times over the past decade.
They're my 2 favorite shooters on the PS3, so it's tough to decide. They both have their strengths - MW2's co-op is a blast, but KZ2's competitive multiplayer is more fun with multiple game modes in one match.
There aren't any FF games on PS3, unless you count the PSOne dowlnoads of FF Tactics, FF7 and FF8. They're all great games and well worth your time and money, but aren't groundbreaking or new by today's standards.
MGS4: skip cutscene and choose the easiest mode, you should be able to beat it in 10 hours.

If you play MGS4 and skip the cutscenes, you're missing the point. If you're just going to skip them then don't even bother playing the game. Seriously.
If you want a game that encourages teamwork, then you might like MAG. If you play it like you would play Modern Warfare, trying to rack up a huge kill/death ratio, then you won't enjoy it. You get the most out of the game when you work with your teammates to complete objectives in the game. If you don't have a headset, then don't even bother. I was in the PRIVATE beta (before the GameStop one) and I had a blast.
If you want a tactical shooter, then MAG is a good choice, but you can't just run & gun like you can in other shooters. Battlefield BC 2 might be better if you want something that you can just pick up and play.

God of War 3 almost took spot #3, but Halo's co-op means it'll get more play-time. Holy crap, it's going to be a great year in 2010.

Of those, Uncharted 2 takes the crown. I still need to finish Assassin's Creed 2 and Ratchet & Clank, but I seriously doubt either will beat out Uncharted.
2.1 Million copies in one month is pretty damn good, especially considering that PS3 has the lowest install base of the three consoles right now. Word of mouth (and a handful of GotY awards) will keep U2: Among Thieves selling for a long time. Anything selling over 1 million is pretty much guaranteed a sequel anyway.
