I think that Joel had hope for the vaccination. However, once he learned that it would cost Ellie her life, his true selfish animal nature caused him to seek after his own goal. In Joel's case I believe that he is trying to restore himself. The world before the infection had obviously not treated him well... Why would he want to bring that back, at the cost of Ellie, and therefore himself?
