Mass Effect 1, I bought a 360 back at xmas '07 just to play that game and boy was it worth it.
A close second would be AC Brotherhood. I played the hell out of the multiplayer on that game, but it was a multiplatform release so I don't know if that counts.

Sure, why not? If someone only owned a 360 they could very well enjoy a multiplat game more than an exclusive one.

Hard pick... but I think I have to go with Bioshock, was the most impressive thing I had ever seen at the time - no game has had that effect on me since.
