I would say yes. If you have xbox live you already have a great deal of what Ninja Gaiden Black has to offer, but still the mission mode is worth getting if you like the game. If you don't have Live, then you definately need Black. New content for every higher difficulty (new enemies replacing old ones or complementing them), a completely new higher difficulty and better balance (e.g. flying swallow isn't the be-all end-all move anymore because most bosses have a counter for it or you can't hit them as easily as before).
Overall it is the better game and the version to get.

Ninja Gaiden Black is well worth it if you like the original. They added new monsters in spots to make it harder such as boss fights and you get the extra content. IMO it's well worth it
