All three. Seriously.. lol
But if you have to choose just one, I'd go with DoS or Lunar Knights.
DoS is easily one of the best 2D sidescrollers, and 2D Castlevania titles, on the DS.. or any system at that. Great gameplay, lots of modes to keep you busy, good cast and an interesting twist to the classic Castlevania story.
LK is really great, though, too. Its a lot like LoZ, but with a 2.5D isotropic view of the action, also, it has a great story and characters, stylized action, fun gameplay and that Hideo flare make this game a great one.
Finally, FFIII is a good choice, if you want a good portable RPG. It's a lengthy game, great graphics, classic (somewhat ancient) RPG gameplay with a decent story.
But, like I said.. out of the 3, DoS and LK both are tied for my top picks.. LK maaaybe having a slight edge, the cutscenes, voice acting and action really are awesome.
