The crossbow from Half-Life 2. Sure, the bolts didn't travel as quick as a bullet, it only holds a single shot at a time (offset by the quick reload)and arcs a bit, but it's almost silent unlike most sniper rifles (or anything meant for long distance shooting) and damn it's fun to nail someone to the wall. Oh yeah, and one shot kills no matter where you hit makes it even more awesome.
Dragon Tooth from Deus Ex. The ultimate melee weapon that isn't a lightsaber. With master level low-tech skill (though it's clearly high tech) and level 4 combat strength, it could take down humanoid opponents in one hit and with some skill, break down locks and doors, takes out cameras and alarms, and destroys large robots with a bit of patience, skill, and a lack of anyone to watch it's back. It sounds game-destroying but on Realistic, you need every trick in the book you can get since enemies can kill you in one hit if you're caught unaware.
The crowbar from Half-Life 1/2. Who needs a voice when it can do all the talking for you? How do you think Gordon got a jobat Black Mesa in the first place?
