It is determined if a solder is psi-gifted the moment you put him or her into the psi-lab as I tested this myself more out of curiosity but can confirm it. What I basically did was this:
1 - Save Game
2 - Put 1 soldier into the Psi-lab
3 - Saved again with quick-save to distinguish these two saves.
4 - Advanced the game 10 days, ignoring all alarms and attacks
5 - Checked the psi-lab report
6 - If Soldier was psi-gifted I just reloaded the Quick-save, if he was not then I reloaded my last regular save and went back to up to 2 and did it all over again.
Be aware that I never did this with Psi-armor on so I do not know if that bonus carries along with the soldiers will or not. But since higher will makes it more likely that he/she is Psi-gifted, using a suit on the soldier you put in the psi-lab can't really hurt as it will most certainly not lower his or her chances.

The mind shield is not a boost to Will, only a boost to Will when defending against an attack.
The in game text suggests that a unit with higher Will is more likely to be gifted, but not guaranteed as I put 2 soldiers in together (both captains) and the one with slightly lower Will was the gifted one. One possibility is to give that unit you know will fail another few missions to rank up - get a higher Will and then test him then.
I don't know if retesting at a later date will have any difference. You could try putting him in for testing on a particular date and advance for 10 days - if it fails, reload, start his testing on the next later date and advance for 10 days. If you keep doing the same test from the exact same time and date it will have the same outcome if the "randomness" is the same as within missions - but a different time and date may work. -
I think whether a soldier is gifted or not is either chosen when they go into testing, or when you first hire them. Will is an indicator, but not a given. I've had three soldiers with will over 80, and only one was gifted, and she wasn't even the highest will. I've also tested squaddies with wills in the mid-40's, and had yet to go on a single mission, that turned out to be gifted.
