Do I have to start a new game on superhero to get the achievement?

Or can i just redo all the missions after changing the difficulty in the settings menu?

The best thing would be to start your first game at superhero difficulty.
When you beat the game you get achievement for beating it at not only super hero but also hero and human difficulty.
But to answer your question i would say yes.
