Just some questions:
Are all the quests completed on the same level every play through?
If I kill Diablo can I play on nightmare/hell mode?
Can any uniques be carried into another game?
How do I get max items?
How can I exploit shrines to level my spells?

No. Some quests are only opened on certain floors, but the majority are between a certain set. Also, on other playthroughs you'll get quests that you didn't encounter the first time. There are of course some mandatory Quests, but you'll know those when you encounter them.

Not directly in Single Player, no. You'll have to host a Multiplayer game to play on the higher skills. (There is a glitch that does allow a SP game to be played in Nightmare/Hell, but I can't remember the details.)
Can any uniques be carried into another game?
Yes, any and all items in your inventory (except certain Quest items, IIRC) are carried across into a new game with that character.
How do I get max items?
Generally just luck, or playing on higher difficulties. There's a certain set of floors which are recommended for 'farming' items, but once again my memory's out to lunch on the matter.
How can I exploit shrines to level my spells?
Save before using the Shrine, and reload the game if it's a crap Shrine I guess. I've never been much of a Spellcasting player so I can't say for sure.

So my single player character is only good for normal difficulty?

So in another topic, called Weapon Advice, I read HegemonKhan say to look up how to play SP nightmare and hell difficulties.
Is there a way?

To be able to play SP on those difficulties, you must first have a MP character that has reached at least level 20, to play Nightmare difficulty, or level 30, to play hell difficulty.
The way the exploit works is to start a MP game at the desired difficulty, and then quit back to the main menu. Next start a SP game, and it will be at the same difficulty as the MP game was.
